Whether you play in the story mode or free play mode, to defeat these characters you've got to do the same thing, which is playing all your notes according to the charts to reach the song's conclusion.

This means that when arrow symbols float and then match above BF, at the same time you need to press the identical arrow keys, hitting the notes.
